Phenacein--an angiotensin-converting enzyme inhibitor produced by a streptomycete. II. Isolation, structure determination and synthesis.

Phenacein, an inhibitor of angiotensin-converting enzyme, has been isolated from the fermentation broth of a Streptomyces species belonging to the Streptomyces tanashiensis-zaomyceticus group. The inhibitor was shown to be 3,6-dihydroxy-1-phenazinecarboxylic acid by spectroscopic, degradative and synthetic methods.

Angiotensin converting enzyme inhibitors produced by Streptomyces chromofuscus. Discovery, taxonomy and fermentation.

Culture A58365.1, NRRL 15098, identified as a new strain of Streptomyces chromofuscus, was found to produce two novel angiotensin converting enzyme (ACE) inhibitors, A58365A and A58365B. Fermentation medium studies afforded an increase in ACE inhibitor titers from less than 1 microgram/ml to greater than 20 micrograms/ml. Proline was the obligatory supplement for ACE inhibitor biosynthesis.

Phenacein--an angiotensin-converting enzyme inhibitor produced by a streptomycete. I. Taxonomy, fermentation and biological properties.

Phanacein, 3,6-dihydroxy-1-phenazinecarboxylic acid, was a specific angiotensin-converting enzyme (ACE) inhibitor isolated from a member of the Streptomyces tanashiensis-zaomyceticus group. Phenacein acted as a pure competitive inhibitor with a Ki of 0.58 microM. ACE inhibition could be reversed by Zn++, but not by Co++, Ca++, or Mg++; therefore, phenacein may chelate the active site zinc of AC...

inflammation3,4). Thus, ICE inhibitors would be useful as anti-inflammatory agents. As described in previous paper5), we isolated novel ICE inhibitory compound, EI-2346, from culture broth of Streptomyces sp. E-2346. In this paper, we describe the structure elucidation of EI-2346. The physico-chemical properties of EI-2346 (1) were summarized in previous paper5). The molecular formula of 1 was ...

K-4, a novel inhibitor of angiotensin I converting enzyme produced by Actinomadura spiculosospora.

A novel inhibitor of angiotensin I converting enzyme (ACE), named K-4, was isolated from the culture broth of Actinomadura spiculosospora nov. sp. K-4. The K-4 was an oligopeptide containing L-phenylalanine with (R)-1-amino-2-(4-hydroxyphenyl)ethylphosphonic acid as the C-terminal residue. The compound proved to be a specific and reversible inhibitor of ACE with the inhibition constant (Ki) of ...
